CloneticsTM 培養基系統（CloneticsTM Media Systems）
培養基說明
CloneticsTM培養基系統是為了使某些特定的正常人體細胞獲得最佳的生長狀態而合成的。可以購買不含添加劑的基礎培養基，也可購買完全培養基，或者便于使用者控制添加劑的種類和濃度的BulleKits方便包裝。每種培養基均檢測了其維持正常人體細胞生長的的能力。每批培養基均經過生化和無菌檢測。
基礎培養基
基礎培養基配方已經經過優化使之適合于某些特定類型的正常人體細胞的培養，不含細胞增殖所必需生長因子。因此必需添加生長因子以加強細胞的接種效率和促進細胞的增殖。
完全培養基
完全培養基是含各種添加劑的生長培養基（BPE和BBE分別各自包裝），含有特定類型正常人體細胞增殖所必須的各種生長因子和添加劑。盡量避免加入成分不確定的添加劑，即便必需也僅使用最小量。所有生長培養基的標準配方都包括抗生素，特殊要求情況下也提供不含抗生素的培養基。
BulletKits
BulletKits提供靈活的終培養基配方及更長的保存期。每個BulletKit含有基礎培養基，已預先測定的專一用途的aliquots(SingleQuots)生長因子和抗生素，便于您根據自己的選擇配置生長培養基。
培養基的制備（Media Preparation）---在無菌環境中執行下列操作。
配制一瓶含各種添加劑的培養基，操作如下：
1， 加BBE或BPE (如需要) 于500ml的基礎培養基瓶中
A,從培養基中分離BBE或BPE添加劑。
B,用乙醇或異丙醇擦凈小瓶和培養基瓶。
C,用吸管將瓶中所有的內容物 (大約2ml) 加入培養基中。用培養基沖洗盛裝BBE的小瓶再將洗液移回500ml瓶中。
D,蓋上蓋子，輕輕地旋轉培養基幾次使之混合。
E,在培養基標簽上標記好BBE或BPE的加入日期。
BulletKits,遵循以下步驟：
1, 用乙醇或異丙醇擦凈SingleQuots 冷凍小瓶和基礎培養基瓶外表面。
2, 無菌條件下打開冷凍小瓶，用吸管將冷凍小瓶中全部內含物加入基礎培養基中。
3, 用培養基清晰每個冷凍小瓶，再將洗液倒回基礎培養基瓶中。要完全達到標示的體積是不太可能的，少量的丟失，即使達到10%通常不會影響含添加劑培養基的細胞生長特性。
4, 我們建議您將試劑盒提供的標簽貼到已加油添加劑的原基礎培養基瓶上，并覆蓋原基礎培養基瓶的標簽，用它來記錄加入的每種添加劑的數量及日期，這樣可避免混淆和可能得雙倍添加。
5, 根據貯藏期限在標簽上注明有效期。完全重新配制的BulletKits 有效期為30天。這種含添加劑的培養基即可以當作一種生長培養基。
注意：考慮到在補充添加劑的過程中可能導致污染，為了確保無菌，整個新配制的生長培養基可能需要使用0.2微米的濾膜重新過濾。由于每次濾過可能導致某些蛋白的丟失，因此我們并不推薦定期的重新濾過。
開始之前---在準備培養基和細胞以前，請遵循以下步驟：
1, 準備一個無菌場地
無菌場地內備有一個帶前置存放開口和濾過層流 級生物安全柜或一個等同的裝置。
2, 決定所需的培養基量
請參閱“常規塑料器皿生長面積表”以此來決定所需的培養基量。
3, 所需的無菌器械和器皿
Ø 無菌可處理的血清學吸液管
Ø 微量移液器和無菌Tips
Ø 可調多通道移液器或重復使用的移液器
Ø 與多通道移液器配合使用的無菌容器
Ø 15ml無菌離心管
Ø 細胞培養瓶
Ø 多孔、平底組織培養板
Ø 血細胞計數儀貨細胞計數器
4, 其他所需物品：
Ø 70%酒精（乙醇或異丙醇）
Ø 生長培養基（細胞類型特異性的）
Ø 防護手套和外套
Ø 臺盼藍
5, 為初始實驗的實施作計劃和準備
依據產品附帶的分析證書上表明的細胞數為依據確定實驗的實施。
6, 核對培養箱的標度。培養箱內應保持一定的濕度并設定為37℃，5%CO2和95%空氣。

Unpacking and Storage Instructions
4. BulletKit® Instructions: Upon arrival, store Basal Medium at 4-8°C and SingleQuots® at -20°C in a freezer that is not self-defrosting. If thawed upon arrival, growth factors can be stored at 4°C and added to basal medium within 72 hours of receipt. After SingleQuots® are added to basal medium, use within one month. Do not re-freeze.
5. ReagentPack™ Subculture Reagents are sterile-filtered and then stored at -20°C until shipment. Subculture reagents may thaw during transport. They may be refrozen once. If you plan to use within 3 days, store at 4°C. Trypsin/EDTA Solution has a limited shelf life or activation at 4°C. If, upon arrival, Trypsin/EDTA is thawed, immediately aliquot and refreeze at –20°C. We recommend that the HEPES-BSS and the Trypsin Neutralizing Solution be stored at 4°C for no more than one month. Note: To keep Trypsin/EDTA fresh and active after thawing, you may aliquot it into sterile centrifuge tubes and re-freeze at -20°C.
Preparation of Medium（Perform the following steps in a sterile field. ）
For EGM® Complete, do the following:
1. Decontaminate the vial of BBE and medium bottle with ethanol or isopropanol.
2. Add the entire contents of the vial (approximately 2 ml) to the media with a pipette. Rinse the BBE vial with medium and pipette the contents back into the 500 ml bottle.
3. Replace the cap and swirl the medium gently a few times to mix.
4. Record the date the BBE was added on the medium label.
For a BulletKit®, perform the following steps:
1. Decontaminate the external surfaces of all supplement vials and the medium bottle with ethanol or isopropanol.
2. Aseptically open each supplement vial and add the entire amount to the basal medium with a pipette.
3. Rinse each cryovial with the medium. It may not be possible to recover the entire volume listed for each cryovial. Small losses, even up to 10%, should not affect the cell growth characteristics of the supplemented medium.
4. Transfer the label provided with each kit to the basal medium bottle being supplemented. Use it to record the date and amount of each supplement added. We recommend that you place the completed label over the basal medium label (Avoid covering the basal medium lot # and expiration date) to avoid confusion or possible double supplementation.
5. Record the new expiration date on the label based on the shelf life.
Note: If there is concern that sterility was compromised during the supplementation process, the entire newly prepared growth medium may be refiltered with a 0.2 μm filter to assure sterility. Routine refiltration is not recommended.
